I think this moment is actually really telling. Loki isn’t frightened of mortals and their “impressive” weapons. He is (more or less) immortal and can’t be seriously injured by whatever they cook up to subdue him. That’s why he’s so confident of his plan to rule them. “An ant has no quarrel with a boot.” etc. etc.
However, let’s introduce the newest addition to the super secret boyband party. Immortal, one of the most powerful beings in the cosmos, and pissed off as all hell. If I was sitting where Loki is, I’d be near shitting my pants. He hasn’t seen Thor since his fall from the bifrost, and for all he knows, Thor’s about to take Odin’s role as his primary torturer (because that’s basically Loki’s mindset towards the Allfather at this point) and intends to bring him back to face “justice” for all the things Loki’s done in the time between Thor and The Avengers.
I like to think a lot of time has passed between then and now. Because it seems to me time on Asgard passes way faster than on Earth. Thor says “In my youth, I called it war.” when speaking to Coulson, which makes me think at least several centuries have passed for him. Centuries during which he’s mourned for Loki and wished desperately to have him back.
But yeah. He’s pissed as all hell here because his brother’s been abducted by these foolish mortals and how dare they take the one person who means more to him than the nine realms combined.
